QMTT Cell Viability Assay Kit

由美商 BioChain 公司所出品，藉由 MTT 化合物可經由活細胞粒線體脫氫酶反應，還原為紫色結晶物質的特性，偵測細胞存活率或生長速率。比一般 MTT 試劑更加靈敏，僅需 950 個細胞即可偵測得訊號。

HEK293 cells were plated in clear bottom 96-well plates. Cells were serial diluted from 55,000 cells to 25 cells in 100 mL DMEM plus 10% FBS. 15 mL of the QMTT reagent w as added and cells were incubated for 4 hours at 37°C. Then 100 mL of the Solubilization Solution was added and the plate was shaken for 1 hour at room temperature. The OD570nm (cm-1) was measured on a Molecular Devices SpectraMax 384 Plus reader. A linear relationship was observed between OD570nm and the cell number. The detection limit was estimated to be 950 cells from the blank control.   Cytotoxic effects of saponin in HEK293 cells. HEK293 cells were plated at 55,000 cells per 80 mL well in clear bottom 96-well plate. Cells w ith treated with varying concentrations of saponin. After overnight incubation, 15 mL of QMTT reagent was added and further incubated for 4 hours. 100 mL of the Solubilization Solution w as then added. The plate was shaken for 1 hour at room temperature and OD570nm (cm-1) was read on a Molecular Devices SpectraMax 384 Plus reader. The IC50 for saponin was 0.0026 wt%.
