GrowDex® animal-free hydrogels

GrowDex® 是新一代 Hydrogel (水膠或稱為水凝膠),僅由來自木材的奈米纖維素 (nanofibrillar cellulose) 及純水組成,組成單純,完全無動物成分,可以模擬細胞外基質 (ECM) 提供細胞生長與分化的支撐。

GrowDex® 水凝膠為即用型凝膠,穩定性佳,可在室溫保存、運輸和使用,提供方便快捷的使用體驗。GrowDex® 已被許多期刊文獻使用在癌細胞、初代細胞、幹細胞及組織類器官 (organoids )、球狀體 (spheroids) 的 3D 細胞培養實驗,應用於癌症治療、藥物篩選及再生醫學等研究。

相較於含動物成分的 Matrigel, GrowDex® 能夠維持更佳的批間穩定性及高再現性的實驗結果,並避免動物成分造成的免疫反應,同時可讓形成的球狀體 (spheroids) 大小更一致,因此已逐漸發展為 3D 細胞培養技術的新研究模式。綜合其優勢特點包含:

相較於 Matrigel,在 GrowDex 培養出的腫瘤球更趨近球形,且細胞形態也更均勻一致

Following 96h of culture in either GrowDex or Matrigel, HepG2 cells were seen to form spheroids throughout the hydrogel. Microscopy images (Nikon Eclipse, 10x) showed that the HepG2 cells formed into spheroids when embedded in the GrowDex hydrogel. These spheroids had a more spherical and homogenous morphology. On the other hand, spheroids formed in Matrigel showed random shaped morphology, uncontrolled growth, and spheroid merging.

相較於 Matrigel,在 GrowDex 培養的細胞無論是在不同 well 或不同批次產品測試中,皆展現出更穩定一致的細胞生長狀態,打下良好的實驗再現性基礎

The cell viability ratio of HepG2 cells was calculated from 96h/24h viabilities in GrowDex and Matrigel. Data is shown as a scatter plot with the Mean ± SD measured from cells cultured in three different lots of hydrogel, with 5 repeat wells per lot, repeated 3 times. There was little variation between test sample replicates and lots when cells were cultured in GrowDex. On the other hand, there was a lot of variation in the viability ratio of test sample replicates and lots for cells cultured in Matrigel.
